The Variable Expression of TGF-¥â1 and TNF¥á Correlated with Transglutaminase 1 and 2 expression in Human Head and Neck Squamous Cell Carcinoma Cell Lines(HNSCC)


Abstract
Human head and neck squamous cell carcinomas(HNSCCs) are a major tumor of oral and pharyngeal cancer. It is well known that TGase(Transglutaminase) is proposed to be related either with defferentiation or with cellular remodeling during invasion and metastasis of malignant tumors. The purpose of this study was to analyze expression level of TGases and various cytokines with 3 cases of HNSCCs from primary and metastatic regions to investigate whether regulation of TGase expression could be related with cytokines. TGase actvity assay, TGase protein determination by immnoslot bloting and mRNA determination by semi-quantitative RT-PCR were processed in cultured normal human oral keratinocyte (NHOK), immortalized human oral keratinocyte(IHOK) and HNSCC cell lines. From the aboving results, the elevated expression of TNF¥á was correlated with the increased TGase 2 expression in HNSCCs, while the increased expression of TGase 1 was correlated with the elevated expression of TGF-¥â1 in cultured NHOK abd IHOK. It was suggested that the increased expression of TGF-¥â1 would be related to the differentiation of NHOK and IHOK by THase 1, while the elevated expression of TNF¥á would be to antiproliferation of HNSCCs by TGase 2.
